Hello,

I looked up SOE (splicing by overlap extension) in order to ligate two DNA fragments to each other with PCR.

I fully understand the technique, although I`m still struggling with the design of the tail of the reverse primer (of the first fragment) and the forward primer (of the second). How long should these tails be? And what about the sequence itself? For the reverse primer (of the first fragment), should it be the start of the second fragment, the reverse of that sequence or its reverse complement? And what about the forward primer of the second fragment?
